2.1 Classification of the substance or mixture
Note This chemical does not meet GHS hazard criteria for 83.6% (534 of 639) of all reports. scheme(5), an estimated BCF of 3.2(SRC), from an estimated log Kow of 0.75(6) and a regression- derived equation(7), suggests the potential for bioconcentration in aquatic organisms is low(SRC). The hydrolysis of 3-methacryloxypropyl-trimethodxysilane proceeds within a few minutes in a low pH environment as a result of the catalytic action of H+ ions(8). Biodegradation data were not available(SRC, 2009). Recycle any unused portion of the material for its approved use or return it to the manufacturer or supplier. Ultimate disposal of the chemical must consider: the material's impact on air quality; potential migration in soil or water; effects on animal and plant life; and conformance with environmental and public health regulations. Data Bank (HSDB) 11.4.3 Preventive Measures ... Hazard is the generation of considerable exothermic heat in some of the reactions, so that high pressures & temp may develop. This danger ... should be borne in mind when designing plant. Awareness of the dangers and of good engineering design are essential to safety. Employees should be instructed about the necessity of cleansing the skin if it is contaminated by materials which are irritants or skin-absorbed. With careful design, however, and complete enclosure of those processes where toxic chemicals or intermediates occur, dangerous exposures can be avoided. / Acrylic acid & derivatives/ International Labour Office. Groups of 10 mice (5 male and 5 female) received a single dose of 2500, 4000, 5000 mg/kg body weight of test article in corn oil; blood samples were drawn 30, 48 and 72 hours after injection. A statistically significant increase in the incidence of micronucleated polychromatic erythrocytes was not observed (Fischer exact test, p > 0.001) at any dose level or time of sampling. A statistically significant (linear regression analysis, r = -0.97) dose related decrease in the PCE/NCE ratio was observed at the 72 hour sampling time (all doses), reflecting the toxic effect of the test article. Additionally, in a preliminary toxicity test, administration of 5000 mg/kg test article (maximum testable dose) induced a 25% decrease in the polychromatic to normochromatic erythrocyte (PCE/NCE) ratio, while mortality was not observed at doses up to and including this level. Bushy Run Research Center; Silane A-174: In Vivo Mouse Micronucleus Study (1986), EPA Document No. FYI-OTS-0386-0469, Fiche No. OTS0000469-0 Data Bank (HSDB) The frequency of chromosomal aberrations was determined in Chinese Hamster Ovary (CHO) cells exposed to 2- propionic acid, 2-methyl, 3-(trimethoxysilyl)propyl ester (silane A-174) with and without metabolic activation provided by Aroclor-induced rat liver S9 fraction. CHO-K1-BH4 cells were exposed to test article at concentrations ranging from 0.1 to 0.6 mg/ml in the presence and absence of activation, and samples were withdrawn at 8 and 12 hours for evaluation. In activated tests, statistically significant (Fischer's exact test, p < 0.05) increases in the incidences of aberrations were observed in samples withdrawn at 8 hours from cultures exposed to 0.3, 0.4, and 0.6 mg/ml, and in samples withdrawn at 12 hours from cultures exposed to 0.2 and 0.3 mg/ml. Additionally, it was reported that the frequency of sister chromatid exchange was increased in the 12 hour samples . A statistically significant (p < 0.01) increase in the incidence of aberrations was observed in samples withdrawn at 8 hours from nonactivated cultures exposed to 0.4 mg/ml. In preliminary range finding tests, exposure of cells to test article at a concentration of 0.5 mg/ml with activation reduced the percentage of mitotic cells by 62.1%, while exposure to 0.6 mg/ml without activation resulted in a 44.4% decrease. Bushy Run Research Center; Silane A-174: In Vitro Cytogenetic Testing with Chinese Hamster Ovary Cells (1985), EPA Document No. FYI-OTS-0386-0469, Fiche No. OTS0000469-0 Data Bank (HSDB) The ability of methacryloxypropyltrimethoxysilane (MPTS) to induce mutations at the HGPRT gene locus in Chinese Hamster Ovary (CHO) cells in vitro was evaluated in the presence and absence of Aroclor-induced rat liver S9 metabolic activation. Based on preliminary toxicity determinations, MPTS was tested for mutagenicity at concentrations of 0.2, 0.4, 0.6, 0.7 or 0.8 mg/ml without activation and at concentrations of 0.1, 0.2, 0.4, 0.6, 0.7 or 0.8 mg/ml with activation. Cell survival ranged from 108 - 156% without activation and from 42 - 105% with activation. There were no significant differences between treated and control (methanol solvent vehicle) cultures with respect to mutation frequencies at any of the concentrations tested, either with or without activation. Acidic conditions enhanced the hydrolysis and the ensuing silanol entities were quite stable; in fact, these conditions slowed down the rate of the self condensation reactions(1). At high and very low pH values, the rate of hydrolysis is higher than at the neutral pH, at which silanes are most stable; the hydrolysis of 3- methacryloxypropyl-trimethoxysilane proceeds within a few minutes in a low pH environment as a result of the catalytic action of H+ ions(2). ECOLOGICAL INFORMATION
12.2.1 Environmental Fate / Exposure Summary Trimethoxysilylpropyl methacrylate's production and use as an adhesion or coupling agent to dental porcelain, ceramic, or glass surfaces may result in its release to the environment through various waste streams. If released to air, an estimated vapor pressure of 0.44 mm Hg at 25 °C indicates trimethoxysilylpropyl methacrylate will exist solely as a vapor in the atmosphere. Vapor-phase trimethoxysilylpropyl methacrylate will be degraded in the atmosphere by reaction with photochemically-produced hydroxyl radicals; the half-life for this reaction in air is estimated to be 15 hours. The atmospheric half-life for the vapor-phase reaction of trimethoxysilylpropyl methacrylate with ozone has been estimated as 24 hours. Trimethoxysilylpropyl methacrylate does not contain chromophores that absorb at wavelengths >290 nm and therefore is not expected to be susceptible to direct photolysis by sunlight. If released to soil, trimethoxysilylpropyl methacrylate is expected to have low mobility based upon an estimated Koc of 1,700. Volatilization from moist soil surfaces is not expected to be an important fate process based upon an estimated Henry's Law constant of 3.0X10-7 atm-cu m/mole. Trimethoxysilylpropyl methacrylate is not expected to volatilize from dry soil surfaces based upon its vapor pressure. Biodegradation data were not available. If released into water, trimethoxysilylpropyl methacrylate is expected to adsorb to suspended solids and sediment based upon the estimated Koc. Volatilization from water surfaces is not expected to be an important fate process based upon this compound's estimated Henry's Law constant. An estimated BCF of 3.2 suggests the potential for bioconcentration in aquatic organisms is low. The hydrolysis of 3-methacryloxypropyl-trimethodxysilane proceeds within a few minutes in a low pH environment as a result of the catalytic action of H+ ions. A base-catalyzed second-order hydrolysis rate constant of 2.7X10-3 L/mole-sec was estimated using a structure estimation method; this corresponds to half-lives of 81 and 8.1 years at pH values of 7 and 8, respectively. Occupational exposure to trimethoxysilylpropyl methacrylate may occur through inhalation and dermal contact with this compound at workplaces where trimethoxysilylpropyl methacrylate is produced or used. Monitoring data indicate that the general population may be exposed to trimethoxysilylpropyl methacrylate by ingestion or dermal contact during porcelain dental repair work. (SRC)
